Western Blot Shows Human Optineurin Specificity Using Knockout Cell Line.

Western blot shows lysates of U2OS and Optineurin knockout U2OS cell line (KO). Nitrocellulose membrane was probed with Optineurin Antibody (Catalog # NBP3-19900) O/N at 4C, followed by HRP-conjugated Secondary Antibody and ECL detection. A specific band was detected for Optineurin (as indicated) in the parental U2OS cell line, but is not detectable in knockout U2OS cell line. Primary antibody dilution used: 1/1000. The Ponceau stained transfers of each blot are shown. Image, protocol and testing courtesy of YCharOS Inc. (ycharos.com).

Background: Optineurin

Optineurin encodes the coiled-coil containing protein optineurin. Optineurin may play a role in normal-tension glaucoma and adult-onset primary open angle glaucoma. Optineurin interacts with adenovirus E3-14.7K protein and may utilize tumor necrosis factor-alpha or Fas-ligand pathways to mediate apoptosis, inflammation or vasoconstriction. Optineurin may also function in cellular morphogenesis and membrane trafficking, vesicle trafficking, and transcription activation through its interactions with the RAB8, huntingtin, and transcription factor IIIA proteins. Alternative splicing results in multiple transcript variants encoding the same protein. [provided by RefSeq]
