Flow Cytometry: OX40 Ligand/TNFSF4 Antibody (R4930 (Oxelumab)) - Chimeric [NBP2-52687] - Human peripheral blood leukocytes were fixed using 2% PFA, permeabilised using 0.5% Triton and stained with unimmunized rabbit IgG antibody (isotype control, black line) or the rabbit IgG-chimeric version of R4930 (NBP2-52687, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

This antibody is Cytof ready. This chimeric rabbit antibody was made using the variable domain sequences of the original Human IgG1 format, for improved compatibility with existing reagents, assays and techniques. This antibody binds to the co-stimulatory OX40 Ligand/TNFSF4, inhibiting its interaction with OX40.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: OX40 Ligand/TNFSF4

CD252 is a 35 kD member of the TNF superfamily also known as OX40L and CD134L. The OX40 ligand is expressed on activated B cells and antigen presenting cells. OX40 ligand interacts with OX40 antigen (CD134) expressed predominantly on activated T cells to increase proliferation and IL-2 production, and to enhance proliferation and immunoglobulin secretion in activated B cells. The RM134L antibody can inhibit OX40/OX40 ligand interactions and block the costimulatory activity of OX40L.
