Western Blot: PAK1 [p Ser204] Antibody [NBP2-29665] - Untreated and pervanadate-treated NIH/3T3 cells (lanes 1 and 2, respectively) were resolved by electrophoresis, transferred to PVDF and probed with anti-phospho-PAK1 (Ser204) (1:1,000). Blocking Peptide Analysis: Pervanadate-treated NIH/3T3 lysate was either unblocked, phosphorylated-peptide blocked, or non-phosphorylated peptide blocked (lanes 3, 4, and 5, respectively) that are that were resolved by electrophoresis, transferred to PVDF and probed with anti-phospho-PAK1 (Ser204) (1:1,000). Lanes 4 and 5 were blocking peptides were the phosphorylated peptide used as the immunogen and the non-phopshrylated form of the peptide. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates phosphorylated PAK1.

Background: PAK1

p21-activated kinases (PAK) are Ser/Thr kinases, which are activated by, and are effectors of Rho family of GTPases. As such, they are implicated in Actin polymerization and activation of the stress-activated kinase cascades. PAK1 is phosphorylated by cdk5, resulting in its inhibition. PAK2 can be activated by Caspase-cleavage in addition to p21-binding, implicating it in cytoskeletal changes during apoptosis. PAK3 is expressed at high levels in post-mitotic neurons of the developing post-natal cerebral cortex and hippocampus, and is the mutant gene thought responsible for non-syndromic X-linked mental retardation.
