Cells were fixed with 4% PFA, permeabilzed with 0.3% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-Human PARK7 monoclonal antibody (1:60) at 37? 1 hour. Then cells were stained with the Alexa Fluor® 594-conjugated Goat Anti-rabbit IgG secondary antibody (red) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

Background: Park7/DJ-1

PARK7, also known as DJ-1, is a member of the peptidase C56 protein family. The PARK-7 protein is a positive regulator of androgen receptor-dependent transcription. It has also been reported to be a potential sensor for oxidative stress, a redox-sensitive chaperone, and a key protein for preventing neuronal oxidative stress and cell death.

PARK7 is highly expressed in pancreas, kidney, skeletal muscle, liver, testis and heart, and is detected at slightly lower levels in placenta and brain. The protein is also detected in astrocytes, Sertoli cells, spermatogonia, spermatids and spermatozoa.

Defects in this gene cause the autosomal recessive neurodegenerative disorder early-onset Parkinson's disease 7.
