Cytoplasm. Nucleus. Mitochondrion. Note: Under normal conditions, located predominantly in the cytoplasm and, to a lesser extent, in the nucleus and mitochondrion. Translocates to the mitochondrion and subsequently to the nucleus in response to oxidative stress and exerts an increased cytoprotective effect against oxidative damage. Detected in tau inclusions in brains from neurodegenerative disease patients.

Background: Park7/DJ-1

PARK7, also known as DJ-1, is a member of the peptidase C56 protein family. The PARK-7 protein is a positive regulator of androgen receptor-dependent transcription. It has also been reported to be a potential sensor for oxidative stress, a redox-sensitive chaperone, and a key protein for preventing neuronal oxidative stress and cell death.

PARK7 is highly expressed in pancreas, kidney, skeletal muscle, liver, testis and heart, and is detected at slightly lower levels in placenta and brain. The protein is also detected in astrocytes, Sertoli cells, spermatogonia, spermatids and spermatozoa.

Defects in this gene cause the autosomal recessive neurodegenerative disorder early-onset Parkinson's disease 7.
