Flow Cytometry: PCNA Antibody (PC10) [DyLight 488] [NB500-106G] - An intracellular stain was performed on HepG2 cells with PCNA Antibody (PC10) NB500-106G (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 10 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to DyLight 488.

Background: PCNA

PCNA, also known as polymerase delta auxiliary protein, is essential for DNA replication and is involved in DNA excision and mismatch repair pathways. PCNA binds to the CDK inhibitor p21, the structure-specific endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT). PCNA is a potentially useful marker of cells with proliferative potential and for identifying the proliferation status of tumour tissue (i.e. relevant to prognosis).
