Western Blot: PCNA Antibody (SY12-07) [NBP2-67390] - Analysis of PCNA on L6 cell lysates with Rabbit anti-PCNA antibody at 1/500 dilution. Lysates/proteins at 10 ug/Lane. Predicted band size: 29 kDa Observed band size: 36 kDa Exposure time: 2 minutes; 12% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Background: PCNA

Proliferating Cell Nuclear Antigen (PCNA) is a DNA polymerase that is essential for DNA replication, excision and mismatch repair pathways. PCNA is a "DNA clamp" which binds to the CDK inhibitor p21, the structure-specific endonucleases Fen1 and XPG, and DNA cytosine 5-methyltransferase (MCMT). PCNA is a potentially useful marker of cells with proliferative potential and for identifying the proliferation status of tumor tissue (i.e. relevant to prognosis). PCNA antibodies are also useful tools for DNA repair and cancer research.
