This Pma1 Antibody (40B7) is useful for Immunocytochemistry/Immunofluorescence and Western blot analysis. For Western blots of yeast protein samples (cell lysates), use at a dilution of 1:10,000, followed by chemiluminescent detection (ECL). For non-ECL western detection methods, use at a dilution of 1:1,000 to 1:5,000. The predicted molecular weight by western blot is 100 kDa.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Pma1

Pma1 (plasma membrane H+-ATPase) belongs to cation transport ATPase (P-type) (TC 3.A.3) family and is an essential protein in yeast/Saccharomyces cerevisiae wherein it pumps out H+ for controlling intracellular pH as well as for generating an electrochemical gradient required to drive cellular import of metabolites (this activity utilizes cellular ATP). Glucose acts as an inducer of Pma1 expression and its catalytic activity through phosphorylation at Ser-899, Thr-912 and Ser-911 sites. Upon glucose starvation, Pma1 is negatively regulated by casein kinase I (Yck1p and Yck2p) phosphorylation and glucose mediated Pma1 activation requires glucose sensor Snf3p, G-protein Gpa2p and transport/phosphorylation of glucose. Hsp30 negatively regulate Pma1 activity during heat shock conditions and contributes towards thermo-tolerance by lowering cellular ATP consumption. Pma1 expression has been shown to induce oncogenic transformation of NIH 3T3 cells and cause increased intracellular pH. Pma1 has emerged as a potential target for development of anti-fungal agents, its suitability as a biomarker for toxicity mediated stress and as a model for study of membrane protein biogenesis.
