Background: POT1

POT1 (protection of telomeres protein 1), a member of telombin family, is expressed ubiquitously and localizes to nucleus/chromosomal telomeres. It forms a component of telomerase RNP (ribonucleoprotein) complex which is essential for chromosome termini replication. POT1 participates in shelterin complex/telosome (contains TERF1, TERF2, TINF2, TERF2IP, ACD and POT1 - regulates telomeric length alongwith protecting telomers), telomerase holoenzyme complex (TERT, DKC1, WRAP53/TCAB1, NOP10, NHP2, GAR1, TEP1, EST1A, POT1 and TERC) and is also found complexed with TERF1, TINF2 and TNKS1. POT1 interacts with TNKS1 and can heterodimerize with ACD for enhancing telomere elongation via increase in telomerase processivity. POT1 implicates in double-stranded telomeric DNA-binding TRF1 complex which is essential to the regulation of telomere length by cis-inhibition of telomerase. POT1 acts as a single-stranded telomeric DNA-binding protein and thus behaves as a downstream effector of TRF1 complex and may transduce information about telomere maintenance and/or length to the telomere terminus. POT1 functional alterations can lead to G-strand overhang loss, chromosomal-end fusions, chromosomal rearrangements, and rapid cell cycle arrest, ultimately leading to senescence and apoptosis. Accordingly, POT1 expression is altered in tumors and is specifically upregulated in therapeutic/radiation-resistant cancer cells.
