Product Summary for Progesterone R/NR3C3 Antibody (SPM566)

Immunogen

Recombinant full-length human Progesterone R/NR3C3 protein (PGR) (Uniprot: P06401 )

Localization

Nuclear

Specificity

This monoclonal antibody is specific to progesterone receptor and shows minimal cross-reaction with other members of the family. Progesterone receptor is expressed as two major isoforms, PR-A (81kDa) and PR-B (116kDa). Expression of PgR has been suggested to reflect a intact estrogen regulatory machinery and therefore, predict better clinical response to endocrine therapy than ER alone. It is excellent for immunohistochemical staining of formalin/paraffin tissues.

Application Notes
Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
Optimal dilution for a specific application should be determined.
Please Note: Optimal dilutions of this antibody should be experimentally determined.

Background: Progesterone R/NR3C3

The human Progesterone Receptor (PgR) is a member of the steroid family of nuclear receptors. PgR is found as a 120 kDa protein (Form B) or a 94 kDa protein (Form A) due to the use of alternative translation initiation sites. In its inactive state, PgR forms a multiprotein complex which includes heat shock proteins and immunophins. Upon binding of progesterone hormone to its receptor, there is a conformational change that allows dimerization and binding of the receptor to progesterone response elements (PRE) sequences, resulting in activated transcription.
