Chromatin Immunoprecipitation: RAR alpha/NR1B1 Antibody [NBP3-11869] - ChIP assays were performed using NB4 cells, NBP3-11869 and optimized primer pairs for qPCR. Sheared chromatin from 6 million cells and 4 ul of antibody were used per ChIP experiment. QPCR was performed using primers specific for the TGM2, HMHA1, PRAM1 and H2B genes. This image shows the relative occupancy, calculated as the ratio + control/background for which the second exon of the MB gene was used.

The plates were coated with the peptides used for immunization of the rabbit. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1:2,400.

Background: RAR alpha/NR1B1

Retinoic acid, a metabolite of vitamin A, is necessary for normal organogenesis but acts as a teratogen at high levels during embryonic and fetal development. Retinoic acid functions through its interaction with the nuclear protein, retinoic acid receptor (RAR). RAR belongs to the steroid and thyroid hormone superfamily of nuclear receptor proteins which exert their effects by binding to specific DNA response elements, thus regulating gene expression in target cells. RAR exists as three major subtypes: alpha, beta and gamma.
