Immunohistochemistry: Rictor Antibody [NB100-1534] - The combined silencing of Rictor and EGFR in vivo results in a complete inhibition of tumor growth. U251Ng2x, U251Rictor, U251EGFR and U251EGFR/Rictor cells were implanted into the right caudate nucleus-putamen of Rag2M mice. Induction of shRNA expression in mice was initiated on day 21 by dissolving 2 mg/mL doxycyline and 5% sucrose in drinking water. On day 49, animals were imaged by Maestro (TM) fluorescence imaging unit expression of tRFP co-expressed with the shRNA sequences. Mice brains were harvested, sectioned and stained for nuclei, Rictor, EGFR and p(473)-AKT and imaged for all markers in addition to tRFP by robotic fluorescence microscopy. No tumor was detected in the U251EGFR/Rictor group. Application Notes
WB: Preliminary experiments gave an approx. 38 kDa band in human lung carcinoma A549 cell line and cervix epitheloid carcinoma cell line HeLa lysates after 0.1 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 192 kDa band according to NP_689969.2. The 38 kDa band was successfully blocked by incubation with the immunizing peptide. IHC-P: kidney shows strong cytoplasmic staining of epithelial cells in distal convoluted tubules.

Background: Rictor

RICTOR is a component of a protein complex [mTORC2] that integrates nutrient- and growth factor-derived signals to regulate cell growth. Rictor is a key component of mTORC2 which plays an essential role in Akt phosphorylation and signaling. Deregulation of Akt/protein kinase B (PKB) is implicated in the pathogenesis of cancer and diabetes. Rictor-mTOR may serve as a drug target in tumors that have lost the expression of PTEN, a tumor suppressor that opposes Akt/PKB activation.
