Background: SCP1

SCP1 (synaptonemal complex protein 1), also called cancer/testis antigen 8 (CT8), belongs to a growing family of cancer/testis antigens (CTA) which includes several proteins (MAGE, GAGE/PAGE/XAGE, NY-ESO-1/LAGE-1, SSX, SPANX, TRAG-3, BAGE, OY-TES-1, CT17, NY-BR-3 and SCP-1) that are not expressed in normal tissues except for testis and, in some cases, placenta. SCP1 is found in a complex with SYCE1 and SYCE2, and is a major component of the transverse filaments of synaptonemal complexes. SCP-1 is established to be expressed selectively during the meiotic prophase of spermatocytes formation and is involved in the pairing of homologous chromosomes during meiosis I. SCP1 is localized in tripartite segments of synaptonemal complexes, between lateral elements in the nucleus, and is found only where the chromosome cores are synapsed. Its N-terminus is found towards the center, whereas, the C-terminus extends well into the lateral domain of the synaptonemal complex. SCP-1 differs from other members of CTA family by its localization on chromosome 1 and it is frequently observed to have variable expression in neoplastic tissues/cell lines including malignant gliomas, breast, renal cell, ovarian, pancreatic and hepatocellular carcinomas. Because of exclusive expression of SCP1 in testis/placenta, it is emerging as an important target for cancer research and therapy.
