Flow Cytometry: SIRP beta 1/gamma Antibody (OX117) - Chimeric [NBP2-81072] - Flow-cytometry using [NBP2-81072]. Human lymphocytes were stained with an isotype control (NBP2-52638, panel A) or the rabbit-chimeric version of OX117 (NBP2-81072, panel B) at a concentration of 1 ug/ml for 30 mins at RT. After washing, bound antibody was detected using a AF488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques. This antibody binds to human Signal-regulatory protein gamma, an immunoglobulin-like cell surface receptor binding CD47. It mediates cell-cell adhesion and enhances T-cell activation.

Background: SIRP beta 1/gamma

The protein encoded by this gene is a member of the signal-regulatory-protein (SIRP) family, and also belongs to the immunoglobulin superfamily. SIRP family members are receptor-type transmembrane glycoproteins known to be involved in the negative regulation of receptor tyrosine kinase-coupled signaling processes. This protein was found to interact with TYROBP/DAP12, a protein bearing immunoreceptor tyrosine-based activation motifs. This protein was also reported to participate in the recruitment of tyrosine kinase SYK. Multiple transcript variants encoding different isoforms have been found for this gene.
