Immunised mice and constructed IgG1 Fab phage display libraries from the spleen cells. SLC7A5/LAT1-reactive recombinant Fab fragments were retrieved by epitope peptide panning.

Epitope

Shown to bind to epitopes of sequences; LLHGDFHAFSAG-GGGC-NH2, LLHGDFH-GGGC-HN2, and FHAFSAG-GGGC-NH2.

Specificity

Shows no reactivity to live HeLa cells by ICC/IF.

Clonality

Monoclonal

Host

Mouse

Isotype

IgG1 kappa

Description

CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(IM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background: SLC7A5/LAT1

Mammalian cells have several amino acid transport systems. These systems have been defined by both activity and specific genes. L-type amino acid transporter 1 (LAT1) is a 12 membrane-spanning protein. LAT1 is a Na + -independent neutral amino acid transporter agency and essential for the transporter of large neutral amino acid such as Leucine, Isoleucine, and Valine through the plasma membrane. LAT1 has been proposed to be one of the major nutrient transport systems at the blood-brain barrier. Drugs such as L Dopa are transported by LAT1. LAT1 is thought to be up-regulated to support the high protein synthesis for cell growth in some tumor cell lines.
