Immunohistochemistry-Paraffin: Smad7 Antibody - Azide Free [NB100-56688] - IHC analysis of a formalin fixed paraffin-embedded (FFPE) human liver using Smad7 antibody on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Cytoplasmic staining of Smad7 was observed. Staining was performed by Histowiz. Image using the standard format of this product.

Background: Smad7

SMADs are members of the MAD-related family of molecules. MAD-related proteins are a family of intracellular proteins that are essential components in the signaling pathways of the serine/threonine kinase receptors of the transforming growth factor beta superfamily (1). SMADs can be divided into receptor-regulated SMADs (R-SMADs: SMAD1, 2, 5, 8 and 9), common-mediator SMAD (co-SMAD: SMAD4), and inhibitory SMADs (I-SMADs: SMAD6 and SMAD7). SMAD1, SMAD5, SMAD8 and SMAD9 have high degrees of homology and antibodies are available that recognize sequences common to all of them. SMAD8 and SMAD9 are typically used as alternate names for one another in the literature.
