This antibody may have a broad banding pattern, typical of glycosylated proteins, at ~35-40 kDa. One may see multiple bands migrating between 35-65 kDa. Do not boil the sample prior to loading on the gel for Western Blot (60 degrees Celsius appears to work fine).

Background: Sodium Potassium ATPase Beta 1

Sodium Potassium ATPase Beta 1 (NKA-beta1), member of X(+)/potassium ATPases subunit beta family, is a single-pass type II membrane protein (transmembrane enzyme) responsible for active reciprocal transport of Na+/K+ ions across plasma membranes, and it contains a larger alpha subunit which catalyze ATP hydrolysis and a smaller beta subunit which is the non-catalytic component of the active enzyme. The alpha subunit (113 kD) contains the binding sites for ATP, Na+ ions, K+ ions, and allosteric sites for inhibitors and activators, and is capable of catalyzing ATP hydrolysis as an essential energy source to transport 3 Na+ ions out of the cell and 2 K+ ions into the cell against membrane ion gradients. The beta subunit (35 kD) is a glycoprotein with isoforms beta1, beta2 and beta3 which are non-catalytic in nature because they lack binding sites for ATP or Na+/K+ ions. In fact, beta subunit is a specific chaperone that assists the biogenesis and corrects membrane insertion of newly synthesized NKA. Moreover, it exerts allosteric activities and by altering the apparent affinity of Na+ /K+ ions as well as by enhancing enzyme turnover rate, NKA-beta1 remotely controls the catalytic function of the enzyme through JY2948-beta1-alpha1 interaction complex. Furthermore, NKA-beta1 may dictate functional NKA concentrations in caveolae, can promote translation efficiency of the alpha subunit and mediate alpha/beta assembly as well as beta-beta homo-oligomerization, and is essential to epithelial polarization, suppression of invasion, and cell motility.
