Background: Somatostatin R1/SSTR1

Somatostatin is a growth hormone inhibitory peptide that regulates diverse cellular functions such as neurotransmission, cell proliferation, and endocrine signaling as well as inhibiting the release of many hormones and other secretory proteins. Somatostatins act through a family of related G protein coupled receptors that are variably expressed in a wide variety of tissues and tumors (kharmate G, 2011). Five genes encoding six different somatostatin receptor subtypes (SSTR1, SSTR2A, SSTR2B, SSTR3, SSTR4, and SSTR5) have been cloned. These genes are intron-less and code for a protein approximately 400 amino acids in size. SSTR1 is widely expressed in pituitary and gastroenteropancreatic tumor, renal, colorectal and breast cancer, meningioma, glioma neuroblastoma, and pheochromocytoma (Barbieri F, 2013).After SST binding to SSTR1 via a protein, JAk2 is activated causing, in turn, phosphorylation of SHP-2. The activation of SHP-2 induces Src dephosphorylation at the inhibitory site, followed by its activation via phosphorylation of the tyrosine 418. Finally, active Src directly induces PTP phosphorylation and activation, being this event the final effector of SST antiproliferative activity. SSTR1 also inhibits adenylyl cyclase when expressed in heterologous CHO-k1 cells (Arena S, 2007). SSTR1 robustly activates the MAP kinase and augments bFGF-induced stimulation of ERk. Activation of ERk via SSTR1 is mediated by the; subunit of a pertussis toxin-sensitive G-protein and requires both Ras and Raf-1. Moreover the phosphatidyl inositol-3 kinase and the cytosolic tyrosine kinase c-src participate in the signal transduction regulated by SSTR1 to activate ERk. SSTR1 Antibody recognizes endogenous levels of total SSTR1 protein. The presence of SSTR1 in tumor cells indicates a potential for somatostatin to bind and suppress growth, as well as allowing for therapeutic treatment with somatostatin analogues (Carless MA, 2001).
