Immunocytochemistry/Immunofluorescence: Syndecan-4 Antibody (047) [NBP2-90700] - Immunofluorescence staining of SDC4 in Hela cells or mouse splenocytes. Cells were fixed with 4% PFA, permeabilzed with 0.1% Triton X-100 in PBS, blocked with 10% serum, and incubated with rabbit anti-mouse SDC4 monoclonal antibody (1:60) at 4C overnight. Then cells were stained with the Alexa Fluor(R) 488-conjugated Goat Anti-rabbit IgG secondary antibody (green) and counterstained with DAPI (blue). Positive staining was localized to cytoplasm.

Background: Syndecan-4

Syndecans are type I integral membrane proteoglycans that are involved in cell-extracellular matrix adhesion and growth factor binding. Syndecan-1 is an matrix receptor which binds to Collagens, Fibronectin and Thrombospondin. Syndecan-1 and Syndecan-3 interact with MK (midkine), a growth/differentiation factor involved in embryogenesis. Syndecan-2 is highly expressed at areas of high morphogenetic activity, such as epithelial-mesenchymal interfaces and the prechondrogenic and preosteogenic mesenchymal condensations. Syndecan-4 functions cooperatively with integrins in the processes of cell spreading and focal adhesion assembly.
