Western Blot Shows Syntaxin-BP1 Specificity Using Knockout Cell Line.

Western blot shows lysates of U-87 MG human glioblastoma/astrocytoma parental cell line and Syntaxin-BP1 knockout U-87 MG cell line (KO). Nitrocellulose membrane was probed with Syntaxin-BP1 Antibody (S05-4B6) (Catalog # NBP3-15097) followed by HRP-conjugated secondary antibody. A specific band was detected for Syntaxin-BP1 at approximately 68 kDa (as indicated) in the parental U-87 MG cell line, but is not detectable in knockout U-87 MG cell line. Primary antibody dilution used: 1/2000. The Ponceau stained transfer of the blot is shown. This experiment was conducted under reducing conditions. U-87 MG human glioblastoma/astrocytoma cell line lysates were prepared and immunoprecipitation was performed using 2.0 μg of Syntaxin-BP1 Antibody (S05-4B6) (Catalog # NBP3-15097) pre-coupled to Dynabeads Protein A. Immunoprecipitated Syntaxin-BP1 was detected in Western Blot with NBP3-15097 used at 1/2000. The Ponceau stained transfer of the blot is shown. Background: Syntaxin-BP1

Munc-18 (mammalian homologue of Unc-18) is a protein that is thought to be involved in regulating exocytosis due, at least in part to its ability to bind syntaxin (Ciufo et al., 2005). Munc18-1 is a neuron-specific member of the Sec1/Munc18 protein family that binds to syntaxin1A and is thought to stabilize the complex (Liu et al., 2004). The function of Munc-18 is thought to be regulated by PKC phosphorylation of Ser515 on the Munc-18 protein (Sassa et al., 1996).
