Application Notes
This antibody is Cytof ready. This full-length, reformatted mouse antibody was made using the variable domain sequences of the original Mouse scFv format, for improved compatibility with existing reagents, assays and techniques. The antibody binds specifically to the recombinant and native variable domain of the alpha chain which comprises the T-cell receptor (TCR). The TCR is a disulfide-linked heterodimeric protein expressed on the surface of T-cells in complex with CD3 and zeta-chain molecules. The receptor functions in the recognition of MHC molecule and the subsequent activation of T-cells. The monoclonal antibody 34 recognises V-alpha regions of TCRs expressed in the 85.33 arthritogenic T-cell hybridoma and can be used to analyse TCR V gene expression and for immunotherapy in T cell mediated autoimmunity.

Background: TCR alpha

T cell receptor (TCR) is a heterodimer consisting of an 945; and a 946; chain (TCR alpha/beta) or a 947; and a delta chain (TCR gamma/delta). TCR-beta is a member of the immunoglobulin superfamily and a component of the CD3/TCR complex (along with TCR-alpha). It is expressed on alpah/beta TCR-bearing T cells and thymocytes. The CD3/TCR complex plays a key role in antigen recognition, signal transduction, and T cell activation. The MAB0872 antibody does not cross-react with delta/gamma TCR-bearing T cells.
