Product Specifications

Immunogen

mAb desire-1 was obtained after fusion of spleen cells from a BALB.K mouse immunised twice with KB5-C20 clone cells in RPMI 1640 medium, with non-Ig-producing myeloma cells X63-Ag8.653. Desire-1 was selected for its capacity to inhibit selectively the H-2Kb-specific cytolysis by clone KB5-C20.

Specificity

mAb desire-1 recognises an epitope contributed by both the alpha and beta chains of the KB5-C20 TCR.

Clonality

Monoclonal

Immunocytochemistry/Immunofluorescence: TCR alpha/beta Antibody (Desire-1) - Chimeric [NBP3-09021] - Immunofluorescence analysis of paraformaldehyde fixed mouse splenocytes immobilized on Shi-fix(TM) cover-slips and stained with the chimeric rabbit IgG version of Desire-1 (NBP3-09021) at 10 ug/ml followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining in a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-09021, DAPI, merged channels and an isotype control. The isotype control was stained with anti-Fluorescein antibody followed by Alexa Fluor(R) 488 secondary antibody.

Applications for TCR alpha/beta Antibody (Desire-1) - Chimeric - Azide and BSA Free

Application
Recommended Usage

Flow Cytometry

Optimal dilutions of this antibody should be experimentally determined.

Immunocytochemistry/ Immunofluorescence

Optimal dilutions of this antibody should be experimentally determined.

Immunoprecipitation

Optimal dilutions of this antibody should be experimentally determined.
Application Notes
This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG format, for improved compatibility with existing reagents, assays and techniques.

Desire-1 has been used for the immunoprecipitation of soluble TCR molecules (Gregoire 1991 & Hua 1986). An FITC conjugated desire-1 has been used in flow cytometric analysis as a marker for clonotype expression (Schonrich 1991). Further, a Fab fragment derived from desire-1 has been used in the crystallisation of the T-cell receptor for 3D structure determination (Housset 1997).

Background: TCR alpha/beta

The antigen-specific T cell receptor (TCR) is composed of either alpha and beta subunit, or gamma and delta subunit. Majority of T cells present in the blood, lymph and secondary lymphoid organs express TCR alpha/beta heterodimers, whereas the T cells expressing TCR gamma/delta heterodimers are localized mainly in epithelial tissues and at the sites of infection. The subunits of TCR heterodimers are covalently bonded and in the endoplasmic reticulum they associate with CD3 subunits to form functional TCR-CD3 complex. Lack of expression of any of the chains is sufficient to stop cell surface expression.
