Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-USP1 antibody NB100-88117 used for IP at 6 ug per reaction. USP1 was also immunoprecipitated by rabbit anti-USP1 antibodies NB100-88116 and NB100-88118. For blotting immunoprecipitated USP1, NB100-88117 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: USP1

This gene encodes a member of the ubiquitin-specific processing (UBP) family of proteases that is a deubiquitinating enzyme (DUB) with His and Cys domains. This protein is located in the cytoplasm and cleaves the ubiquitin moiety from ubiquitin-fused precursors and ubiquitinylated proteins. The protein specifically deubiquitinates a protein in the Fanconi anemia (FA) DNA repair pathway. Alternate transcriptional splice variants have been characterized. [provided by RefSeq]
