Product Summary for VAV2 [p Tyr142] Antibody

Immunogen

Phospho-Vav2 (Tyr-142) synthetic peptide (coupled to KLH) corresponding to amino acid residues surrounding Tyr-142 in mouse Vav2. This sequence has high homology with similar regions in rat and human Vav2, and has significant homology to the conserved site in Vav1 and Vav3. [UniProt# Q60992]

Specificity

This antibody was cross-adsorbed to unphosphorylated Vav2 (Tyr-142) peptide before affinity purification using phospho-Vav2 (Tyr-142) peptide (without carrier). The antibody detects a 100 kDa band corresponding to Vav on SDS-PAGE immunoblots of human A431 cells treated with EGF and detects Vav1, Vav2, and Vav3 in Jurkat cells treated with pervanadate. The antibody does not detect these Vavs after treatment with alkaline phosphatase.

Immunocytochemistry/Immunofluorescence: VAV2 [p Tyr142] Antibody [NBP3-23268] - Immunocytochemical labeling of VAV2 in control and pervanadate-treated human A431 cells. The cells were fixed in paraformaldehyde and permeabilized using NP-40. Then labeled with rabbit polyclonal VAV2 (Tyr-142). The antibody was detected using goat anti-rabbit DyLight(R) 594.

Background: VAV2

The Vav family are Rho/Rac guanosine nucleotide exchange factors (GEFs), consisting of three members in mammalian cells (Vav, Vav2, Vav3) and one in nematodes (CelVav) (1). Vav and Vav2 are members of the dbl family of guanine nucleotide exchange factors (GEF) for the rho/rac family of GTP binding proteins. Vav is expressed primarily in hematopoietic cells, while vav2 has a wider tissue distribution (2). Vav2 serves as a guanine nucleotide exchange factor for the Rho family of Ras-related GTPases. Unlike Vav1, whose expression is restricted to cells of hematopoietic origin, Vav2 is broadly expressed. Recently, Vav2 has been identified as a substrate for the epidermal growth factor (EGF) receptor. EGF receptor phosphorylates Vav2 on all three possible phosphorylation sites, Tyr-142, Tyr-159, and Tyr-172. results suggest that EGF regulates Vav2 activity basically through phosphatidylinositol 3-kinase activation, whereas tyrosine phosphorylation of Vav2 may rather be necessary for mediating protein-protein interactions (3)
