Product Summary for Human IL-1 alpha/IL-1F1 Quantikine ELISA Kit

The Quantikine Human IL-1 alpha Immunoassay is a 3.5 or 4.5 hour solid phase ELISA designed to measure IL-1 alpha in cell culture supernates, serum, and plasma. It contains recombinant human IL-1 alpha and antibodies raised against recombinant human IL-1 alpha and has been shown to accurately quantitate the recombinant factor. Results obtained using natural IL-1 alpha showed linear curves that were parallel to the standard curves obtained using the E. coli-expressed Quantikine kit standards. These results indicate that this kit can be used to determine relative mass values for natural human IL-1 alpha.

Sample Values

Serum/Plasma - Forty samples from apparently healthy volunteers were evaluated for the presence of human IL-1 alpha, and all had levels which fell below the lowest IL-1 alpha standard, 3.9 pg/mL. No medical histories were available for the donors used in this study.

Cell Culture Supernates - Human peripheral blood mononuclear cells (5 x 106 cells/mL) were cultured in RPMI supplemented with 5% fetal bovine serum, 50 μM beta-mercaptoethanol, 2 mM L-glutamine, 100 U/mL penicillin, and 100 μg/mL streptomycin sulfate. Cells were cultured unstimulated or stimulated with 10 μg/mL PHA. Aliquots of the cell culture supernates were removed on days 1 and 5 and assayed for levels of human IL-1 alpha.

Background: IL-1 alpha/IL-1F1

IL-1 alpha (Interleukin-1 alpha) is a cytokine that is induced during microbial exposure and promotes the release of multiple inflammatory mediators and acute phase proteins. It also supports hematopoietic stem cell expansion, promotes neutrophil mobilization from the bone marrow, and promotes osteoclast formation. IL-1 alpha signals through a receptor complex composed of IL-1 RI and IL-1 RAcP. It also binds the decoy receptor IL-1 RII which associates with IL-1 RAcP but does not transduce signaling.
