Product Summary for Mouse IFN-beta Quantikine ELISA Kit

The Quantikine® Mouse IFN-beta Immunoassay is a 4.5 hour solid phase ELISA designed to measure mouse IFN-beta in cell culture supernates, serum, and plasma. It contains
HEK293-expressed recombinant mouse IFN-beta and antibodies raised against the recombinant factor. This immunoassay has been shown to quantitate the recombinant factor accurately. Results obtained using natural mouse IFN-beta showed dose-response curves that were parallel to the standard curves obtained using the recombinant kit standards. These results indicate that this kit can be used to determine relative mass values for natural mouse IFN-beta.

Sample Values

Serum/Plasma - Ten serum samples, 5 EDTA plasma, and 5 heparin plasma samples were evaluated for the presence of mouse IFN-beta in this assay. No detectable levels were observed.

Cell Culture Supernates - - NIH-3T3 mouse embyronic fibroblast cells were cultured in DMEM supplemented with 10% fetal bovine serum and grown to approximately 90% confluency. Cells were unstimulated or stimulated with 10 μg/mL Poly I:C delivered via Lipofectamine for 24 hours. Aliquots of the cell culture supernates were removed and assayed for levels of mouse IFN-beta.

Precision

Intra-Assay Precision (Precision within an assay) Three samples of known concentration were tested twenty times on one plate to assess intra-assay precision.

Inter-Assay Precision (Precision between assays) Three samples of known concentration were tested in twenty separate assays to assess inter-assay precision. Assays were performed by at least three technicians using two lots of components.

Linearity

To assess the linearity of the assay, samples spiked with high concentrations of mouse IFN-beta were serially diluted with calibrator diluent to produce samples with values within the dynamic range of the assay.

Background: IFN-beta

Interferon-beta (IFN-beta) is a cytokine that is produced by fibroblasts and pathogen-exposed dendritic cells, macrophages, and endothelial cells. It signals through the heterodimeric IFN-alpha/beta Receptor. IFN-beta-deficient mice show increased susceptibility to experimental autoimmune encephalomyelitis (EAE), a disease model of human multiple sclerosis (MS). Furthermore, IFN-beta has been shown to suppress the Th17 cell response in both MS and EAE and has commonly been used as a treatment for MS.
