Description

The stop solution changes color from blue to yellow and the intensity of the color is measured at 450 nm using a spectrophotometer. In order to measure the concentration of Human PARP in the sample, this Human PARP ELISA Kit includes a set of calibration standards. The calibration standards are assayed at the same time as the samples and allow the operator to produce a standard curve of optical density versus Human PARP concentration. The concentration of the samples is then determined by comparing the O.D. of the samples to the standard curve.

Background: PARP

Poly (ADP-ribose) polymerase (PARP) is protein family primarily invloved in DNA repair and programmed cell death. PARP proteins are activated in response to DNA single strand breaks (SSBs). Upon SSB detection, PARP binds the DNA and synthesizes a PAR chain at the damaged site, which then signals the initiation of DNA repair by other enzymes such as XRCC1, DNA ligase III and DNA polymerase beta. After repair, the PAR chains are degraded via PAR glycohydrolase.

PARP is inactivated by caspase cleavage, leading to a programmed cell death pathway.
