This lyophilized preparation is stable at 2-8 degrees C, but should be kept at -20 degrees C for long term storage, preferably desiccated. Upon reconstitution, the preparation is most stable at -20 to -80 degrees C, and can be stored for one week at 2-8 degrees C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degrees C to -80 degrees C. Avoid repeated freeze/thaw cycles.

Purity

> 97 % pure by SDS-PAGE and HPLC

Endotoxin Level

Less than 0.1 EU/ug of CX3CL1/Fractalkine as determined by LAL method.

Predicted Molecular Mass

8.7 kDa.
Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Activity

CX3CL1 Protein is fully biologically active when compared to standard. The ED50 as determined by a cell proliferation assay using human peripheral blood lymphocytes (PBL) is less than 0.5 ug/ml, corresponding to a specific activity of > 2000 IU/mg.

Background: CX3CL1/Fractalkine

CX3CL1 recently identified through bioinformatics is the only known member of the CX3C chemokine family and it is also commonly known under the names fractalkine (in humans) and neurotactin (in mice). Unlike other known chemokines, CX3CL1 is a type 1 membrane protein containing a chemokine domain tethered on a long mucinlike talk. The soluble form of CX3CL1 is chemotactic for T-cells and monocytes, but not for neutrophils. CX3CL1 may play a role in regulating leukocyte adhesion and migration processes at the endothelium. Mouse CX3CL1 shares 85 % and 78 % a.a. sequence identity with rat and human CX3CL1, respectively, within the chemokine domain, but lower sequence identity within other domains.
