Activity

Biological activity measured by its ability to inhibit BMP-4-induced alkaline phosphatase production by ATDC5 mouse chondrogenic cells. The ED50 for this effect is 0.04-0.2 ug/mL in the presence of 50 ng/mL of Recombinant Human BMP-4.

Reconstitution Briefly centrifuge vial prior to opening to bring the contents to the bottom. Reconstitute in sterile distilled water or aqueous buffer containing 0.1% BSA to 0.1-1.0 mg/mL. Stock solutions should be apportioned into working aliquots and stored at -20C.
Shipping The product is shipped with polar packs. Upon receipt, store it immediately at the temperature recommended below.
Stability & Storage Store at -20 to -70C as supplied. After reconstitution, store at 2 to 8C for 1 month and at -20 to -70C for long term storage. Avoid repeated freeze-thaw cycles.

Background: Noggin

Noggin is involved in numerous developmental processes, such as neural tube fusion and joint formation. The morphogenesis of organs is initiated by a downgrowth from a layer of epithelial stem cells. This process is achieved through the receipt of signals from 1) a WNT protein (WNT3A) to stabilize beta-catenin; and 2) Noggin, which is a bone morphogenetic protein inhibitor. Noggin mutations in unrelated families with proximal symphalangism (SYM1) and multiple synostoses syndrome (SYNS1) have been identified, which have multiple joint fusion as their principal defect.
