Background: ErbB2/Her2

ErbB2, also called human epidermal growth factor receptor 2 (HER2) and Neu, is a type I membrane glycoprotein and is one of four members of the ErbB family of receptor tyrosine kinases. ErbB family members serve as receptors for the epidermal growth factor (EGF) family of growth factors. Cynomolgus ErbB2 consists of an extracellular domain (ECD) with two ligand binding domains and two cystine rich domains, a transmembranee region and a cytoplasmic domain with a flexible juxtamembrane region followed by a protein kinase domain (1). The ECD of mature cynomolgus ErbB2 shares 99% amino acid sequence identity with the ECD of human ErbB2. ErbB2 signaling is associated with neurodegenerative diseases such as multiple sclerosis and Alzheimer disease when impaired, while over-expression of ErbB2 signaling is associated with development of cancers (2). ErbB2 is widely expressed in epithelial cells and has been primarily associated with breast carcinomas (3, 4). Over-expression of HER-2 also caused other cancers such as gastric (2,5), colorectal (6), ovary, lung, bladder, head and neck (7), and osteosarcoma (8). Among ErbB family members, ErbB2 is unique in that it has no identified ligands. Rather, ErbB2 heterodimerizes with the other members of the ErbB family to form higher affinity signaling complexes (9). ErbB2 can be shed from the cell surface by the protease ADAM10, resulting in a constitutively active truncated intracellular receptor (10). ERBB2 is used as biomarkers in several cancers and is an attractive oncogenic driver and therapeutic target in cancers (6).
