Activity

Recombinant Human His6-UBE2F is a member of the NEDD8-conjugating (E2) enzyme family that receives NEDD8 from a NEDD8-activating (E1) enzyme and subsequently interacts with a NEDD8 ligase (E3) to conjugate NEDD8 to substrate proteins. Reaction conditions will need to be optimized for each specific application. We recommend an initial Recombinant Human His6-UBE2F concentration of 0.1-1 μM.

Background: UBE2F

Ubiquitin-conjugating Enzyme E2F (UBE2F) is a NEDD8-conjugating enzyme (E2) (1). It is a 185 amino acid (aa) protein with a predicted molecular weight of 21 kDa. The human protein exhibits approximately 96% aa sequence identity with its mouse and rat orthologs. UBE2F is involved the covalent attachment of NEDD8 to target proteins in a process termed neddylation. For example, UBE2F appears to participate in the neddyylation of Cullin 5 (1). Members of the Cullin family (Cullin 1-7) are scaffold proteins that can incorporate into multimolecular complexes termed Cullin RING Ubiquitin Ligases (CRLs) (2). CRLs make up the largest subset of Ubiquitin Ligases (E3), and their activity is promoted by neddylation of associated Cullin family proteins (3).
