Background: Plexin B1

Plexin B1, also known as SEP, is a type I transmembrane glycoprotein that belongs to the Plexin family of semaphorin receptors. In humans, the Plexin family consists of at least nine members divided into four subfamilies and Plexin signaling is involved in neuronal axon guidance, angiogenesis and immune response (1). Plexins share a common structural architecture: A large extracellular domain (ECD) that binds the semaphorin ligand, a single transmembrane domain and a conserved ~600 amino acid (aa) cytoplasmic region (2, 3). Human Plexin B1 contains one Semaphorin domain, two plexin-semaphorin-integrin (PSI) domains, and three immunoglobin-plexin-transcription factor (IPT) repeats in the ECD. The mature ECD of human Plexin B1 shares 82% aa sequence identity with the ECD of mouse Plexin B1. Like semaphorins, plexins can be soluble or membrane bound and Plexin B1 can be cleaved into an ~200 kDa, soluble alpha-subunit and an ~100 kDa membrane bound beta-subunit (2). Plexin B1 is a receptor for Semaphorin 4D/CD100 and it can form a receptor complex with Neuropilins, MET, and EGF R2 (4). Plexin B1 is expressed in chick retinal ganglion neurons and hippocampal neurons with a peak during the myelination period (5). Plexin B1 also plays a role in GABAergic development, changes of actin cytoskeleton, axon guidance, cell migration, and invasive growth (6, 7).
