Background: DcTRAILR1/TNFRSF23

Decoy TRAIL receptor 1 (DcTRAIL R1), also called TNFRSF23 is a glycosylphosphatidylinositol (GPI)-anchored cell surface protein that binds TRAIL/TNFSF10 with high affinity (1). Mouse DcTRAIL R1 consists of a 29 amino acid (aa) signal peptide, a 126 aa mature region and a 21 aa GPI anchor signal. The mature region shows a TNFR cysteine rich domain (CRD) with three tandem A1/B2 repeats (2). Compared to signaling TRAIL receptors, DcTRAIL R1 does not trigger death signaling due to its lack of an intracellular death domain. Also, unlike many TNF receptors family members, DcTRAIL R1 does not contain a TRAFF-binding motif. Thus DcTRAIL R1 failed to induce NF kappaB activity. Structural studies revealed a strict species selectivity of the mouse DcTRAIL R1 for mouse and not human TRAIL based on amino acid specific electrostatic interactions between the two proteins (2). Mouse DcTRAIL R1, DcTRAIL R2 (TNFRSF22), and TNFRSF26 are encoded by clustered genes within the Kcnq1 domain of chromosome 7. The human counterpart of the Kcnq1 domain does not encode a functional homolog of the murine DcTRAIL R1 (3). Mouse DcTRAIL R1 is weakly expressed on CD4+CD25+ Tregs but not on CD4+CD25 conventional T cells. The difference in DcTRAIL R1 expression between the two T cell subsets may affect their responsiveness to TRAIL (4).
