Background: Fibulin 7

Fibulin 7, also known as TM14, is an approximately 46 kDa member of the fibulin family of proteins (1, 2). Structurally, Fibulin 7 contains an N-terminal Sushi domain, 3 central tandem EGF modules, and a C-terminal domain homologous to the fibulin module (2). Mouse Fibulin 7 mRNA is highly expressed in teeth, with weaker expression in brain, kidneys, muscles, and bones (2). Mouse Fibulin 7 shares 86% amino acid sequence identity with human Fibulin 7. Fibulin 7 interacts with several extracellular molecules, including Fibronectin, DSPP, and Fibulin 1 (2). A C-terminal fragment of Fibulin 7 was able to bind Human Umbilical Vein Endothelial Cells (HUVECs) and inhibit HUVEC tube formation and vessel sprouting in aortic ring assays, suggesting that Fibulin 7 may have anti-angiogenic activity (3). Fibulin 7 may be important for proper development, since it is included in a chromosomal deletion that is associated with severe cardiac defects and craniofacial anomalies (4).
