This lyophilized preparation is stable at 2-8 degrees C, but should be kept at -20 degrees C for long term storage, preferably desiccated. Upon reconstitution, the preparation is most stable at -20 to -80 degrees C, and can be stored for one week at 2-8 degrees C. For maximal stability, apportion the reconstituted preparation into working aliquots and store at -20 degrees C to -80 degrees C. Avoid repeated freeze/thaw cycles.

Purity

> 97 % pure by SDS-PAGE and HPLC

Endotoxin Level

Less than 1 EU/ug of TFF3 as determined by LAL method.

Predicted Molecular Mass

13.2 kDa.
Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Activity

Trefoil Factor 3 protein is fully biologically active when compared to standard. The ED50 as determined by a chemotaxis bioassay using human MCF-7 cells is less than 10 ug/ml, corresponding to a specific activity of > 100 IU/mg.

Background: TFF3

Trefoil Factor 3 (TFF3), also known as ITF and P1.B, plays an important role in epithelial regeneration and wound healing. It is expressed by epithelial goblet cells in the respiratory tract, biliary and breast ducts, small and large intestine, and cardia of the stomach. It is upregulated in response to a range of gastrointestinal epithelial disruptions and induces the migration of biliary, bronchial, and intestinal epithelial cells. Its upregulation is also associated with and enhances tumor cell invasion and metastasis. TFF3 promotes angiogenesis and enhances VEGF upregulation in tumor cells. It can form disulfide-linked dimers or associate into disulfide-linked complexes with other intestinal mucous proteins including FCGBP and MUC-2.
