PURE-EVs are size exclusion chromatography (SEC) columns designed to isolate extracellular vesicles (EVs) from small volume samples. Compared to other techniques, PURE-EV efficiently separates EVs from circulating proteins while maintaining original shape and functionality of vesicles. Highly pure EV preparations, no residual chemicals from precipitation, and rapid isolation in approximately 20 minutes. Isolated EVs are suitable for a wide range of downstream analyses such as nanotracking analysis (NTA), protein profiling (western blotting, ELISA, Flow Cytometry), and nucleic acid extraction.

EV SEC Columns [NBP3-11763]

EV SEC Columns [NBP3-11763] - Results and EV separation. EV SEC column was filled with 2 ml of medium from HCT116 cells, concentrated 10 folds by Tangential Flow Filtration (TFF)-EV concentrator (Catalog # NBP3-11762). 24 fractions (500 μl each one) have been collected and analyzed by ELISA assay (CD81 marker) and by BCA test for determining respectively vesicle and total protein content. EVs are eluted in fractions 8 - 12 (turnaround time approximately 15 min), whereas circulating proteins corresponded to the fractions 14 - 24.

Background: EV SEC Columns

Size-exclusion chromatography (SEC) separates macromolecules based on their size. To accomplish this, the sample is passed through a porous column and molecules are isolated in fractions from the eluate. The size of the pores can vary depending on the size of the desired product. For isolation of EVs from proteins and lipoproteins, pore sizes can vary from ~40-80 nm. As the sample passes through the column, molecules smaller than the pore size will get trapped in the pores, slowing them down, while larger molecules will pass through. This results in the larger molecules, such as EVs, eluting from the column in earlier fractions, while smaller molecules, such as proteins and protein aggregates, are eluted in later fractions.
