PAM functions as an E3 ubiquitin ligase for tuberin (TSC2), a negative regulator of the mTOR pathway. Additionally PAM has been shown to function as a guanosine exchange factor (GEF) for Rheb and to facilitate sphingosine-1-phosphate signaling in the mTOR pathway.
