TORC3, also known as CREB-regulated transcription coactivator 3 (CRTC3), is a 67 kDa protein that functions as a coactivator for CREB1 in promoting transcription through both consensus and variant cAMP response element (CRE) sites. TORC3 activity is important in regulating the expression of genes involved in cellular energy metabolism. Its binding to the HTLV-1 protein Tax enhances HTLV-1 transcription, whereas its binding to cellular Bcl3 inhibits HTLV-1 transcription. Alternate splicing of mouse TORC3 generates isoforms that lack the N-terminal 299 amino acids.
