Immunocytochemistry/Immunofluorescence: Caspase-9 Antibody (8G5) [NBP3-26696] - Staining of HepG2 with Caspase-9 Antibody (8G5) at 1:7, counter-stained with DAPI. The cells were fixed in 4% formaldehyde and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 525-conjugated Goat Anti-Rabbit IgG (H+L).

The cells were fixed in 4% formaldehyde and permeated by 0.2% Triton X-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4C. Control antibody (green line) was rabbit IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background: Caspase-9

Caspase 9 (CASP9) is a member of the cysteine-aspartic acid protease (caspase) family that is involved in the execution-phase of cell apoptosis. Caspase 9 is expressed as an inactive proenzyme which then undergos proteolytic processing by upstream caspases, to form an active (cleaved) caspase 9 form.

There are two classes of caspases involved in apoptosis; initiators (activation by receptor cluster) and effectors (activation by mitochondrial permeability transition). Caspase 3 is considered an initiator, as initiated caspase-9 goes on to cleave the effector procaspase-3 and procaspase-7.

Caspase 9 antibodies are important tools for apoptosis studies and research on the caspase family mediated apoptosis signaling pathway.
