Immunocytochemistry/Immunofluorescence: DNA-PKcs [p Ser2056] Antibody (4A4) [NBP3-26398] - Staining of Hela cells(treated with UV) with DNA-PKcs [p Ser2056] Antibody (4A4) at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4C. The secondary antibody was Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L).

Background: DNA-PKcs

The PRKDC gene encodes the catalytic subunit of a nuclear DNA-dependent serine/threonine protein kinase (DNA-PK). The second component is the autoimmune antigen Ku (MIM 152690), which is encoded by the G22P1 gene on chromosome 22q. On its own, the catalytic subunit of DNA-PK is inactive and relies on the G22P1 component to direct it to the DNA and trigger its kinase activity; PRKDC must be bound to DNA to express its catalytic properties.(supplied by OMIM)
