Western Blot: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666]

Western Blot: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666]

Western Blot: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666] - Total protein from mouse placenta was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocking in 5% non-fat milk in TBST. The membrane was probed with 2 ug/ml anti-CD105 (NB100-77666) in 1% milk/TBST and detected with an anti-rat HRP conjugated secondary antibody using chemiluminescence.

Immunocytochemistry/Immunofluorescence: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666] - Using the DyLight 550 direct conjugate Staining of CD105 (endoglin) in 4T1 tumors after radioimmunotherapy shows CD105 expression (red), blood vessels (green), and nuclei (blue).
Immunohistochemistry-Paraffin: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666]

Immunohistochemistry-Paraffin: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666]

Immunohistochemistry-Paraffin: Endoglin/CD105 Antibody (MJ7/18) [NB100-77666] - Analysis of a FFPE mouse ductus deference using a 1:50 dilution of CD105 antibody (clone MJ7/18) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) was performed using Aperio AT2 (Leica Biosystems). Staining was observed in the pseudostratified columnar epithelial cells. Staining was performed by Histowiz.

Background: Endoglin/CD105

CD105, commonly referred to as Endoglin, ENG, HHT1, and ORW1 is a membrane single-pass type I membrane protein limited to endothelial cells in all tissues except bone marrow. It is a major glycoprotein of vascular endothelium and plays a major role in the regulation of angiogenesis. CD105 acts as a TGF-beta coreceptor and is involved in the TGF-beta/BMP signaling cascade. Mutations involving CD105 have been shown to be involved with hereditary hemorrhagic telangiectasia, as well interruptions of the TGF-beta/BMP pathways leading to some cancers. Immunohistochemistry has shown that CD105 has a very high expression in blood vessels of tumors, making it a potential target that can be used in tumor imaging (PMID: 12773481). Elevated serum levels of CD105 in pregnant women have been determined a possible risk factor for preeclampsia (PMID: 24273904).
