Background: FE65

Fe65 (Protein Fe65 or APBB1) is a neuronal adaptor protein that mediates the assembly of multimolecular complexes through a variety of protein-protein interaction domains: the WW domain, which binds to proline-rich sequences, and two C-terminal phosphotyrosine-binding domains. FE65 is primarily found in CNS and is highly expressed in neurons of hippocampus, cerebellum, thalamus, and brain stem nuclei. Fe65 is localized within cytoplasm or nucleus, although its moderate amounts are tethered to membranes via binding with Alzheimer's disease amyloid precursor protein (APP). FE65 forms stable, transcriptionally active complex with AICD (APP intracellular domain) that follows nuclear translocation of AICD and apoptosis through Tip60. FE65 is functionally linked to cellular motility, morphology and actin dynamics through binding of its WW domain to actin-binding protein Mena. It plays a key role in response to DNA damage by translocating to nucleus and inducing apoptosis through recognizing/binding histone H2AX phosphorylated on 'Tyr-142' (H2AXY142ph) at DSBs, recruiting other pro-apoptosis factors such as MAPK8/JNK1. It is required for histone H4 acetylation DSBs and its ability to specifically bind modified histones and chromatin modifying enzymes such as KAT5/TIP60, probably explains its trancription activation activity. It functions in association with TSHZ3, SET and HDAC factors as a transcriptional repressor that inhibits the expression of CASP4.
