Background: GLI-1

GLI1 is a protein originally isolated in human glioblastoma. The GLI proteins are the effectors of Sonic Hedgehog (Hh) signaling and have been shown to be involved in cell-fate determination, proliferation and patterning in many cell types and most organs during embryo development. GLI1 was originally isolated from a glioma tumour and has been found to be up regulated in many tumors including muscle, brain and skin tumors such as Basal cell carcinoma (BCC). Shh and the Gli genes are normally expressed in hair follicles, and skin tumours expressing GLI1 may arise from hair follicles. The level of GLI1 expression correlates with the tumor grade in bone and soft tissue sarcomas. GLI1 expression can be cytoplasm and or nucleus. The GLI1 protein is tethered in the cytoplasm by binding to SUFU. Activation and translocation of the GLI1 protein to the nucleus is promoted by interaction with STK36. Phosphorylation by ULK3 may promote nuclear localization. Translocation to the nucleus is also promoted by interaction with ZIC1.
