Immunocytochemistry/Immunofluorescence: Glucagon R/GCGR Antibody (hGR-2 F6) [NBP2-81065] - Immunofluorescence staining of fixed HepG2 cells with [NBP2-81065]. Immunofluorescence analysis of paraformaldehyde fixed HepG2 cells on Shi-fix coverslips, permeabilized with 0.15% Triton and stained with the chimeric mouse IgG1 version of hGR-2 F6 [NBP2-81065] at 10 ug/ml for 1h followed by Alexa Fluor 488 secondary antibody (2 ug/ml), showing punctate membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP2-81065, DAPI, merged channels and an isotype control. The isotype control was stained with an unknown specificity antibody followed by Alexa Fluor 488 secondary antibody.

Applications for Glucagon R/GCGR Antibody (hGR-2 F6) - Azide and BSA Free

Application
Recommended Usage

Block/Neutralize

Optimal dilutions of this antibody should be experimentally determined.

Immunocytochemistry/ Immunofluorescence

Optimal dilutions of this antibody should be experimentally determined.
Application Notes
This antibody binds to the human glucaon receptor, a GPCR that is activated by glucagon and stimulates adenylate cyclase. The receptor is involved in glucose homeostasis and regulats glucose production in the liver. The antibody acts as a competitive antagonist.

Background: Glucagon R/GCGR

Glucagon, a pancreatic hormone, functions as an antagonist to insulin, stimulating the conversion of glycogen to glucose and increasing blood sugar levels. GLP-1 functions as a transmitter in the central nervous system, inhibiting feeding and drinking behavior. Both glucagon and GLP-1 function through their specific binding to the glucagon receptor or GLP-1R, respectively. The glucagon receptor shows expression in liver, kidney and adipose tissue. The 56 GLP-1R expression primarily localizes to areas of the hypothalamus involved in feeding behavior. Both receptors and their ligands serve as potential targets for the therapeutic treatment of diabetes.
