Detection of Human Apolipoprotein C‑II/ApoC2 by Western Blot.

Western blot shows lysates of human plasma and small intestine tissue. PVDF membrane was probed with 0.25 µg/mL of Human Apolipoprotein C-II/ApoC2 Biotinylated Antigen Affinity-purified Polyclonal Antibody (Catalog # BAF4497) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for Apolipoprotein C-II/ApoC2 at approximately 8 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 2.

Background: Apolipoprotein C-II/ApoC2

Apolipoprotein C2 (ApoC2/II) is a 8‑9 kDa, secreted member of the Apolipoprotein C2 family of proteins. It is produced by hepatocytes and represents a major component of VLDL particles. It activates lipoprotein lipase and may self-associate to form amyloid-type fibrils. The human ApoC2 precursor is 101 amino acids (aa) in length. It contains a 22 amino acid (aa) signal sequence, followed by a 79 aa ProApoC2 that contains a lipid-binding region (aa 43‑51) and an enzyme interaction site (aa 55‑78). ProApoC2 represents > 90% of circulating ApoC2. In human, limited proteolytic processing occurs with removal of the six aa prosegment (aa 23‑28). This does not affect activity. Human ProApoC2 is 59% aa identical to mouse and dog ProApoC2, respectively.
