Detection of Claudin‑8 in HEK293 Human Cell Line Transfected with Human Claudin-8 by Flow Cytometry.
HEK293 human embryonic kidney cell line transfected with human Claudin-8 was stained with Mouse Anti-Human Claudin-8 Monoclonal Antibody (Catalog # MAB5275, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram), followed by Allophycocyanin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0101B). Caki-2 human clear cell carcinoma epithelial cell line was stained with Mouse Anti-Human Claudin-8 Monoclonal Antibody (Catalog # MAB5275, filled histogram) or isotype control antibody (Catalog # MAB002, open histogram), followed by Phycoerythrin-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # F0102B). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). Background: Claudin-8
Claudin-8 is a 225 amino acid (aa), 25 kDa four-transmembrane protein in the Claudin family of tight junction proteins. It is selectively expressed in tight junctions in the kidney thin descending limb, distal tubule and collecting duct, but is also found in liver and intestinal tight junctions. It is thought to function as an ion barrier protein. The extracellular portions of human Claudin-8 share 86% and 89% aa identity with corresponding portions of mouse and rat Claudin-8, respectively.
