Detection of ICAM-3/CD50 in Human Blood Lymphocytes by Flow Cytometry.
Human peripheral blood lymphocytes were stained with (A) Mouse Anti-Human ICAM-3/CD50 Monoclonal Antibody (Catalog # MAB8131) or (B) Mouse IgG1 Isotype Control (MAB002), followed by Allophycocyanin-conjugated anti-Mouse IgG secondary antibody (F0101B) and Mouse Anti-Human CD3 epsilon PE-conjugated Monoclonal Antibody (FAB100P). Staining was performed using our Staining Membrane-associated Proteins protocol.ICAM-3/CD50 in Human Lymph Node.
ICAM-3/CD50 was detected in immersion fixed paraffin-embedded sections of human lymph node using Mouse Anti-Human ICAM-3/CD50 Monoclonal Antibody (Catalog # MAB8131) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cell surface of lymphocytes. Background: ICAM-3/CD50
Intercellular Adhesion Molecule-3 (ICAM-3; also known as CD50), a member of the immunoglobulin superfamily, binds the leukocyte integrins LFA-1 (CD11a/CD18) and alphad/ beta2. ICAM‑3 is expressed on leukocytes and epidermal Langerhans cells. It may play an important role in T cell stimulation by Langerhans cells. ICAM-3 has not been identified in the mouse.
