Detection of Human LAX1 by Western Blot.
Western blot shows lysates of Jurkat human acute T cell leukemia cell line and K562 human chronic myelogenous leukemia cell line. PVDF membrane was probed with 1 µg/mL of Goat Anti-Human LAX1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4706) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). A specific band was detected for LAX1 at approximately 70 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Detection of LAX1 in Jurkat Human Cell Line by Flow Cytometry.
Jurkat human acute T cell leukemia cell line were stained with Goat Anti-Human LAX1 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4706, filled histogram) or isotype control antibody (Catalog # AB-108-C, open histogram), followed by Phycoerythrin-conjugated Anti-Goat IgG Secondary Antibody (Catalog # F0107). Background: LAX1
LAX1 (Linker for activation of X cells) is a 70 kDa member of a small group of T cell transmembrane (TM) adaptor proteins. It is upregulated on both T and B cells after activation, and likely downmodulates both BCR and TCR signaling. Human LAX1 is 398 amino acids (aa) in length. Although its predicted MW is 44 kDa, it runs anomalously at 70 kDa in SDS-PAGE. It is a type III (no signal sequence) TM protein that contains a 37 aa N-terminal extracellular region and a 340 aa cytoplasmic domain (aa 59‑398). There are two potential isoform variants. One has an alternate start site at Met77, while a second shows a 13 aa substitution for aa 1‑29. Over aa 136‑398, human LAX1 is 47% aa identical to mouse LAX1.
