Cells were stained using the NorthernLights™ 557-conjugated Anti-Rabbit IgG Secondary Antibody (red; Catalog # NL004) and counterstained with DAPI (blue). MICL/CLEC12A in Human Spleen.

MICL/CLEC12A was detected in immersion fixed paraffin-embedded sections of human spleen using Rabbit Anti-Human MICL/CLEC12A Monoclonal Antibody (Catalog # MAB29461) at 3 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Rabbit IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC003). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using Antigen Retrieval Reagent-Basic (Catalog # CTS013). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in lymphocytes. Background: MICL/CLEC12A

Human MICL (myeloid inhibitory C-type lectin-like receptor; also CLL-1 and KLRL-1) is a 75 kDa member of the group V C-type lectin-like receptor family of molecules. Group V molecules have a lectin-type domain that binds non-sugar ligands. MICL is a 265 amino acid (aa) type II transmembrane (TM) glycoprotein that contains a 200 aa extracellular domain (ECD). Multiple isoforms of MICL are reported. One potentially utilizes an alternate start site that adds 10 aa to the cytoplasmic domain. There is one potential soluble form that lacks the TM segment, and two truncated ECD isoforms. Human MICL ECD is 53%, 65%, and 55% aa identical to the ECD in mouse, dog and cow, respectively.
