Scientific Data Images for Human NIK/MAP3K14 Antibody

NIK/MAP3K14 in Human Pancreas.

NIK/MAP3K14 was detected in immersion fixed paraffin-embedded sections of human pancrease using Mouse Anti-Human NIK/MAP3K14 Monoclonal Antibody (Catalog # MAB6888R) at 5 µg/mL for 1 hour at room temperature followed by incubation with the Anti-Mouse IgG VisUCyte™ HRP Polymer Antibody (Catalog # VC001). Before incubation with the primary antibody, tissue was subjected to heat-induced epitope retrieval using VisUCyte Antigen Retrieval Reagent-Basic (Catalog # VCTS021). Tissue was stained using DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to cytoplasm in exocrine and endocrine cells. View our protocol for IHC Staining with VisUCyte HRP Polymer Detection Reagents.

Background: NIK/MAP3K14

NIK (NF kappa B inducing kinase), also called MAP3K14, is a widely expressed 947 amino acid (aa), ~100 kDa cytoplasmic protein of the MAP kinase family. NIK participates in a non-canonical NFkB signaling cascade in which its activity and expression are elevated by TNF and related ligands. NIK activates IKKa/b, which releases NFkB subunits for translocation to the nucleus. Within the region used as an immunogen, human NIK shares 94% and 95% aa sequence identity with mouse and rat NIK, respectively.
